چکیده

At a recent conference (1) in Woods Hole, Massachusetts, investigators (2) met to discuss the nematode Caenorhabditis elegans. This free-living worm may, according to some workers, become the Escherichia coli or at least the bacteriophage T4 of the animal world. Small (about I mm in length) and semitransparent, C. elegans provides for research the advantages of a short life cycle (3 days) and a simple anatomy-it contains about 810 nongonadal nuclei. It is both easy to cultivate, on E. coli as a food source, and convenient for genetic analysis. Its genes are carried on five autosomes and a sex chromosome (X), and it has a genome size about 20 times that of E. coli. It generally reproduces as

Dietary restriction in the nematode Caenorhabditis elegans.

The nematode Caenorhabditis elegans has proved to be an excellent model organism for the study of development and aging. Many aging mutants have been discovered in the past two decades, and much has been discovered about the physiology of long-lived mutants. It therefore seems surprising that dietary restriction (DR) has not been extensively studied using C. elegans. The main reason for this is...

A Biologically Accurate Simulation of the Locomotion of Caenorhabditis elegans

The nematode Caenorhabditis elegans is an important model organism for many areas of biological research including genetics, development, and neurobiology. It is the first organism to have its genome sequenced, complete cell ontogeny determined, and nervous system mapped. With all of the information that is available on this simple organism, C. elegans may also become the first organism to be a...

Large-Scale Screening for Targeted Knockouts in the Caenorhabditis elegans Genome

The nematode Caenorhabditis elegans is a powerful model system to study contemporary biological problems. This system would be even more useful if we had mutations in all the genes of this multicellular metazoan. The combined efforts of the C. elegans Deletion Mutant Consortium and individuals within the worm community are moving us ever closer to this goal. At present, of the 20,377 protein-co...

CRISPR-Based Methods for Caenorhabditis elegans Genome Engineering

The advent of genome editing techniques based on the clustered regularly interspersed short palindromic repeats (CRISPR)-Cas9 system has revolutionized research in the biological sciences. CRISPR is quickly becoming an indispensible experimental tool for researchers using genetic model organisms, including the nematode Caenorhabditis elegans. Here, we provide an overview of CRISPR-based strateg...

WormBase 2007

WormBase (www.wormbase.org) is the major publicly available database of information about Caenorhabditis elegans, an important system for basic biological and biomedical research. Derived from the initial ACeDB database of C. elegans genetic and sequence information, WormBase now includes the genomic, anatomical and functional information about C. elegans, other Caenorhabditis species and other...
